Simplifications: positions are rounded, the ends of the PCR product are treated as not ligatable, enzyme compatibility is by identical sites only, colony numbers are random within realistic ranges, and fragments under about 180 bp run off the bottom of this 1% gel and pieces under 100 bp are lost during gel purification. Plasmid names are fictional; pAT is styled after the classic two-marker vector pBR322.

Simplifications: each reaction follows a smooth logistic amplification model; the reference gene amplifies at 100% efficiency; fluorescence is in arbitrary units; replicate noise is a small random wobble in Ct. The ΔΔCt method assumes both primer pairs have about 100% efficiency.
Problems are randomly generated and checked by a separate solver. Distances are in kilobases (kb) on a 0.1-kb grid; you may also type base pairs. Tm uses the Wallace rule, 2(A + T) + 4(G + C) °C. Fold change uses 2 to the power −ΔΔCt. CRISPR problems use SpCas9 with an NGG PAM.
Your job
Drag a card onto a box, or tap a card and then tap the box's name. Cards are buttons, so Tab and Enter work too.
Drag a step by its grip to move it, or use the up and down buttons. Press Check when the order looks right.
Fictional people, animals and data, simplified for teaching. Not medical, legal or genetic counseling advice; real forensic and parentage testing uses many more markers and strict standards.
Wrong picks are fine here. Each one tells you why.
